Description:
The 22URTI monoclonal antibody reacts with human interleukin (IL)-22, a 20 kDa member of the IL-10 cytokine family secreted primarily by Th17 and NK cells. Other T cells can also produce IL-22. In Th17 cultures, IL-22 expression is induced more by IL-23 than IL-6 or TGF-β, suggesting secretion by more differentiated Th17 cells in vivo. The IL-22 receptor is a heterodimer of IL-10R2 and IL-22R1.
Applications Reported:
This antibody is suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-titrated and validated for intracellular staining of restimulated, Th17-polarized CD4⁺ human PBMCs using the Intracellular Fixation and Permeabilization Buffer Set (Product #88-8824-00) and the BestProtocols® protocol (Protocol A: Two-step protocol for cytoplasmic proteins).
Recommended use: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information
IL-22, also known as IL-10-related T-cell derived inducible factor, is an α-helical cytokine in the IFN-IL-10 family (including IL-19, IL-20, IL-24, IL-26, IL-28, IL-29). It is mainly produced by activated T cells and NK cells. The IL-22 gene is located on chromosome 12q and structurally related to IL-10. IL-22 acts via a heterodimeric receptor complex (IL-22R1 and IL-10R2), activating the JAK-STAT pathway.
A soluble receptor, IL-22BP, antagonizes IL-22 binding and signaling. IL-22 primarily affects nonimmune cells such as epithelial cells, keratinocytes, and hepatocytes, regulating genes linked to innate immunity, wound healing, and inflammation. It is co-expressed with IL-17A/F in Th17 cells but differentially regulated. IL-22 plays a critical role in host defense against extracellular pathogens and promotes tissue repair.
