Product Specific Information

This NJTEN3 monoclonal antibody reacts with the pro-form of mouse IL-1 beta, a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It recognizes only the pro-form (31 kDa) and not the active (17 kDa) cleaved form. Activation of inflammasomes triggers cleavage of pro-IL-1β via caspase-1 activation, leading to secretion through a non-classical pathway. IL-1β signals via IL-1RI, shared with IL-1α, and plays a key role in innate immune responses. Its activity can be inhibited by IL-1RA. Elevated IL-1β levels are linked to chronic inflammatory conditions.

Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Tested using mouse thioglycolate-elicited peritoneal macrophages with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended use ≤0.25 µg/test (100 µL final volume). Optimize antibody titration for best results.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Interleukin-1 beta (IL-1β) is a proinflammatory cytokine produced by monocytes, macrophages, and dendritic cells. Synthesized as a 31 kDa pro-form, it is cleaved into an active 17 kDa form via inflammasome-mediated caspase-1 activation. IL-1β lacks a signal peptide and is secreted through an alternative pathway. It signals through IL-1RI and IL-1RII, shared with IL-1α, inducing proinflammatory cytokines and acute-phase responses. IL-1RA competitively inhibits receptor binding. Elevated IL-1β is associated with chronic inflammation, making IL-1β and IL-1RA targets for therapeutic research.
