Product Specific Information

The JES5-16E3 antibody reacts with mouse interleukin-10 (IL-10), an ~18 kDa cytokine also known as Cytokine Synthesis Inhibitory Factor (CSIF). IL-10 is produced by Th2 cells, B1 cells, macrophages, and keratinocytes. It inhibits Th1 cytokine synthesis and T cell proliferation, while promoting mast cell activation and Th2 response.

Applications Reported:
Used for ELISA, ELISPOT, intracellular flow cytometry, IHC, and neutralization assays.

Applications Tested:
Validated for intracellular staining in cultured mouse splenocytes. Recommended usage ≤0.5 µg/test (100 µL final volume). Optimal titration should be determined empirically.

Target Information

IL-10 is a cytokine mainly produced by monocytes and lymphocytes. It regulates immune responses by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It enhances B cell survival and antibody production, inhibits NF-κB activity, and modulates the JAK-STAT pathway. Mouse knockout studies confirm its role as a key immunoregulator in the intestinal tract. Mutations are linked to HIV-1 susceptibility and rheumatoid arthritis. Associated diseases include Graft-Versus-Host Disease and HIV-1 infection.
