Product Specific Information
For reconstitution, add 100 µL of distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments using this carrier-free antibody, perform an additional desalting step (e.g., Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).
Target Information
Programmed death-ligand 2 (PD-L2, also known as B7-DC) is part of the B7 ligand family within the immunoglobulin superfamily and serves as a ligand for PD-1, along with PD-L1.
PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Expression is upregulated by Th2 cytokine IL-4 and, to a lesser extent, by Th1 cytokines TNF-α and IFN-γ.
Although PD-L2 is generally expressed at lower levels than PD-L1, it has a 2–6 times higher affinity for PD-1. PD-1 and its ligands act as inhibitory immune checkpoints that regulate T-cell activation, proliferation, and cytokine secretion (e.g., IFN-γ, IL-10), maintaining immune homeostasis.
Interestingly, PD-L2 can also stimulate T-cell proliferation and cytokine production independent of PD-1, suggesting the presence of additional receptors. One such receptor, repulsive guidance molecule b (RGMb), interacts with PD-L2 and is involved in respiratory tolerance regulation in the lung through BMP and/or neogenin signaling pathways.
