Product Specific Information

The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha has cytolytic activity against various tumor cells and plays a key role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form. MAb11 is a neutralizing antibody.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Validated by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and the BestProtocols® Protocol A.

MAb11 antibody is pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells. Recommended use is 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test.

488–561 nm excitation; 578 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers and shows a single 17 kDa band under non-reducing SDS-PAGE conditions.

Knockout studies suggest its neuroprotective function, and TNF alpha induces tumor necrosis when injected into tumor-bearing mice. It is involved in immune responses to infections and tumor cell cytolysis, synergistic with interferon-gamma. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
