The monoclonal antibody PAJ-17R recognizes mouse IL-17 receptor A (IL-17RA), also known as CD217.
IL-17RA is a single transmembrane receptor that can homo- or heterodimerize with IL-17RC or IL-17RB. It binds to IL-17A, IL-17F (Th17 lineage cytokines), or IL-17E (IL-25). Ligand binding induces receptor dimerization and recruitment of Act-1 via SEFIR domains, activating TRAF6 and signaling through NFκB and MAPK pathways.

Initial studies showed ubiquitous IL-17RA expression, though not all receptor-expressing cells respond to IL-17A. IL-17RA knockout mice display reduced neutrophil numbers and impaired function when challenged with K. pneumoniae or Candida albicans, suggesting IL-17RA’s critical role in neutrophil development and function.

PAJ-17R staining is abundant on Gr-1^dim and Gr-1^bright bone marrow cells and some F4/80 positive cells, but absent in IL-17RA knockout mice.

Target Information

Interleukin 17A (IL-17A) is a proinflammatory cytokine secreted by activated T-lymphocytes. It induces maturation of CD34⁺ hematopoietic precursors into neutrophils.
IL-17RA is a type I membrane glycoprotein that binds IL-17A and IL-17F with low affinity. IL-17A/IL-17RA interactions are implicated in inflammatory and autoimmune diseases such as rheumatoid arthritis.
Mutations in IL-17RA cause familial candidiasis type 5 (CANDF5), a rare disorder with impaired immune responses and defective fungal clearance.
