For reconstitution, add 100 µL distilled water to obtain a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
DNA-directed RNA polymerase II subunit RPB1 (POLR2A) is a DNA-dependent RNA polymerase responsible for transcribing DNA into RNA using ribonucleoside triphosphates as substrates.
POLR2A is the largest subunit and a catalytic component of RNA polymerase II, synthesizing mRNA precursors and various non-coding RNAs. It forms part of the polymerase active center with the second-largest subunit, contributing to the core element, clamp, and jaws that interact with the DNA template.
Transcription initiation involves positioning the single-stranded DNA template within the active site cleft. The bridging helix of RPB1 facilitates translocation of Pol II by conformational changes during nucleotide addition.
Elongation and transcription regulation are influenced by the phosphorylation status of the C-terminal domain (CTD) of RPB1, which serves as a platform for assembly of factors involved in transcription and mRNA processing.
