Product Specific Information

The 25-D1.16 monoclonal antibody reacts with the ovalbumin-derived peptide SIINFEKL bound to H-2Kb of MHC class I, but not with unbound H-2Kb or irrelevant peptide-bound forms.
It is useful for tracking the quantity and localization of specific antigen-presenting cells (APC) in vivo.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes; ≤0.25 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimal titration recommended.

Fluorochrome Information:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is suitable for cytometers equipped with a violet (405 nm) laser. Verify instrument compatibility.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
BV711 is not compatible with methanol-based fixation.
