Description:
The P67.6 monoclonal antibody reacts with human CD33 (also known as GP67 and P67), a 67 kDa type I transmembrane glycoprotein belonging to the Siglec family. It is highly specific to the hematopoietic compartment and expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.
Applications Reported:
This antibody is reported for use in flow cytometric analysis.
Applications Tested:
Pre-titrated and validated by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.
Fluorochrome Information:
eFluor™ 450 is an alternative to Pacific Blue™. It emits at 445 nm and is excited with the Violet laser (405 nm). Ensure the instrument can detect this fluorochrome.

Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains ITIM motifs that recruit SHP-1 and SHP-2 for signal transduction and receptor regulation. CD33 is expressed on myelomonocytic lineage cells, binds sialic acid residues, and is a therapeutic target for acute myeloid leukemia. It is expressed on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells, and serves as a marker for peripheral monocytes. CD33 helps distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
