Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human PEDF. Anti-Human PEDF-specific antibody was purified by affinity chromatography employing an immobilized Human PEDF matrix.

Sandwich ELISA:
To detect Human PEDF by sandwich ELISA (using 100 µL/well), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human PEDF (500-P263) as a capture antibody, allows the detection of at least 2000–4000 pg/mL of Recombinant Human PEDF.

Western Blot:
To detect Human PEDF by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible development reagents, the detection limit for Recombinant Human PEDF is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

PEDF (Pigment Epithelium-Derived Factor) is a noninhibitory serpin with neurotrophic, anti-angiogenic, and anti-tumorigenic properties. It is a 50 kDa glycoprotein produced and secreted in many tissues throughout the body.
A major component of PEDF’s anti-angiogenic action is the induction of apoptosis in proliferating endothelial cells. PEDF also inhibits angiogenic factors such as VEGF and FGF-2.
Its neuroprotective effects are achieved through suppression of neuronal apoptosis induced by oxidative or excitotoxic stress. PEDF binds to a specific receptor (PEDF-R), a lipase-linked cell membrane receptor, which mediates its signaling functions.
Due to its multifunctional nature, PEDF is considered a potential therapeutic target in vasculature-related neurodegenerative diseases (e.g., AMD, PDR) and various cancers involving angiogenesis.
