Immunogen Phospho-WAVE (Tyr-125) peptide (coupled to KLH) corresponding to amino acid residues surrounding Tyr-125 in human WAVE1. This sequence has high homology with similar regions in rat and mouse WAVE1, and has less than 50% homology to similar regions in the conserved site in WAVE2 (Tyr-124) and WAVE3 (Tyr-125).

Product Specific Information

This antibody was cross-adsorbed to phospho-WAVE (Tyr-150) and unphosphorylated WAVE (Tyr-125) peptides before affinity purification using phospho-WAVE (Tyr-125) peptide.
In western blots, the antibody detects an 80 kDa band corresponding to phosphorylated WAVE in human SYF cSrc transformed cells and mouse macrophages treated with pervanadate, but is not observed in control cells.
These bands can be specifically blocked with phospho-WAVE (Tyr-125) peptide (WX1775), but not phospho-WAVE (Tyr-150) peptide (WX1825).

Target Information

WASP (Wiskott-Aldrich syndrome protein) and N-WASP are downstream effectors of Cdc42 that are implicated in actin polymerization and cytoskeletal organization.
The WASP family also includes VASP (vasodilator-stimulated phosphoprotein) and Mena (mammalian enabled protein), which accumulate at focal adhesions and regulate actin cytoskeleton.
WAVE proteins, related to the WASP family, mediate actin reorganization downstream of Rho family GTPases.
WAVE1 and WAVE2 specifically regulate membrane ruffling by inducing actin filament clusters in response to Rac activation.
These proteins cooperate with the Arp2/3 complex to promote actin filament formation.
WAVE1 (also called SCAR) is primarily expressed in the brain, WAVE2 is widely expressed with high levels in leukocytes, and WAVE3 links receptor kinases to actin.
All family members bind actin through a C-terminal verprolin homology domain.
The WAVE multiprotein complex transduces signals that regulate cell shape, motility, and function.
