Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM. CD11b forms a non-covalent complex with CD18 to create the alphaMbeta2 integrin (Mac-1), which binds CD54 (ICAM-1), C3bi, and fibrinogen.
Mac-1 is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and mouse B-1 cells. M1/70 is cross-reactive to human CD11b and can detect this antigen on human peripheral blood monocytes, granulocytes, and NK cells.
CD11b participates in adhesive cell interactions through its ligands.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometry on mouse bone marrow cells. Recommended ≤0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Fluorochrome Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that forms a heterodimer with CD18 (integrin beta-2). The CD11b/CD18 complex, known as Mac-1 or CR3, functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
CD11b is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. It is a common microglial marker in nervous tissue, with expression increasing during monocyte maturation.
Diseases associated with CD11b dysfunction include systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune conditions.
