Product Specific Information

The human Interleukin-35 (IL-35) protein is a dimeric complex consisting of the IL-12a/p35 subunit (NP_000873.2) and the human EBI3 (NP_005746) protein.
This antibody clone (15K8D10) is specific to the EBI3 subunit of the target protein.
Endotoxin level: < 0.06 EU/µg.

Target Information

EBI3 (Epstein-Barr virus Induced-3) is a secreted glycoprotein belonging to the hematopoietin receptor family.
It plays a key regulatory role in Th2-type immune responses and Th2-mediated tissue inflammation, potentially through iNKT cell regulation.
EBI3 forms heterodimers with p28 and p35 subunits to create IL-27 and IL-35, respectively. IL-27, produced by activated antigen-presenting cells upon TLR ligation, negatively regulates Th17 cell differentiation.
EBI3 expression in dendritic cells is transcriptionally regulated by TLR signaling via MyD88 and NF-κB during innate immune responses.
EBI3 signaling inhibits delayed-type hypersensitivity by suppressing IL-17 and inducing IL-10 production.
It may also regulate tumor metastasis through lung CD8+ T cells, and EBI3 deficiency is associated with reduced Th2 cytokine production and decreased allergic airway inflammation in asthma.
Targeted deletion of EBI3 has been shown to protect mice from lung metastasis.
