Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy establishment and maintenance.
PR exists as a 94 kDa protein (Form A) and a 120 kDa protein (Form B), generated by alternative translation initiation. PR-B is transcriptionally active, regulating genes involved in endometrial maintenance, pregnancy, and ovulation inhibition. PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
Inactive PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational changes enable dimerization and DNA binding to progesterone response elements (PRE), activating transcription.
Mutations in the PGR gene can lead to reproductive abnormalities.
