Target Information
Synapse-Associated Protein 102 (SAP102) is a plasma membrane-associated protein found in synaptic junctions. Like other family members, SAP102 contains three PDZ domains (~90 amino acids each), followed by an SH3 domain and a guanylate kinase homology (GuK) domain. PDZ-domain interactions are believed to contribute to receptor and channel clustering, playing a role in neuronal plasticity.
SAP102 is thought to cluster NMDA receptors and shaker-type potassium channels at synaptic membranes in CNS neurons. Both share a C-terminal sequence motif (T/SXV) that mediates PDZ-domain binding. Other neuronal proteins with this motif (β1 adrenergic receptor, certain serotonin receptors, sodium and potassium channel subunits) may also interact with SAP102. Neuronal nitric oxide synthase (nNOS), though lacking the T/SXV motif, can associate with SAP102 via PDZ–PDZ interactions.
