Product Specific Information
Reconstitution:
Lyophilized human M-CSF should be reconstituted in 20 mM Tris-HCl, pH 8.0 to a concentration of 0.1–1.0 mg/mL.
For extended storage, further dilute in a buffer containing a carrier protein (e.g., 0.1% BSA) and store in working aliquots at -20°C to -80°C.
Storage:
Store lyophilized human M-CSF at 2°C to 8°C, preferably desiccated.
Reconstituted protein should be stored at ≤ -20°C (not in a frost-free freezer).
Minimize freeze-thaw cycles.
Target Information
M-CSF (Macrophage colony-stimulating factor, CSF-1) is an essential survival factor for proliferation and development of monocytes, macrophages, and osteoclast progenitor cells. It also induces VEGF secretion by macrophages, promoting endothelial progenitor cell mobilization and neovascularization.
M-CSF exists in several bioactive isoforms differing in potency and stability. The full-length protein is membrane-spanning and may be cleaved and modified in secretory vesicles. It forms a disulfide-bonded homodimer, processed into glycoprotein or proteoglycan isoforms via chondroitin sulfate modification.
Binding to its receptor c-Fms (CSF-1R/CD115) induces receptor dimerization, internalization, and degradation. Functionally, M-CSF stimulates differentiation of hematopoietic stem cells into monocyte-macrophage populations.
Human M-CSF is active on mouse cells, whereas mouse CSF shows no activity on human cells.
