Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 upregulates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
Phosphorylation at Ser15 is critical for p53 activation and stabilization. Mutations in p53 are common in human cancers and result in loss of tumor suppressor function. p53 also plays roles in aging, oxidative stress, and memory-related processes.
It is expressed at low levels in normal cells but highly in transformed cells, contributing to malignancy. Germline mutations of TP53 are associated with cancer-prone syndromes such as Li-Fraumeni.
