Von Hippel-Lindau syndrome (VHL)은 다양한 악성 및 양성 종양 발생에 관여하는 상염색체 우성 유전성 암 증후군입니다. VHL 유전자의 생식세포 돌연변이는 질환의 유전적 원인이며, 이 유전자는 elongin B, elongin C, cullin-2와 복합체를 형성하여 ubiquitin ligase E3 활성을 가집니다. 해당 단백질은 hypoxia-inducible factor (HIF)의 유비퀴틴화 및 분해에 관여하며, 이는 산소에 의한 유전자 발현 조절의 핵심 전사인자입니다. 또한 RNA polymerase II의 POLR2G/RPB7 서브유닛도 표적 단백질로 보고되었습니다. 다양한 스플라이싱 변이체가 존재하여 서로 다른 isoform을 생성합니다.
