Product Specific Information

Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R). It is a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction via tyrosine kinase pathways (Jak/STAT). IL-7 is critical for lymphocyte development and homeostatic proliferation of peripheral T cells, and influences TCR locus rearrangement in γδ T cells.

CD127 expression is down-regulated on CD4+CD25+ regulatory T cells (Tregs). CD4+CD25+ cells with low CD127 expression are enriched for Foxp3+ Tregs with suppressive activity.

Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Each product includes:

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells at 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Product #C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails or single-color controls.
• NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD127 (Interleukin-7, IL-7) is a glycoprotein regulating lymphopoiesis. The active IL7R receptor is an alpha/gamma heterodimer.

• Gamma chain (γc): Activates signal transduction and associates with IL-2 receptor.
• Alpha chain (IL7Rα): Determines specific signaling events via cytoplasmic molecules.
  CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
  Dysfunction of CD127 is associated with severe combined immunodeficiency (SCID).
