Target Information

The pax genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, enabling them to regulate gene expression.

Several pax genes are associated with developmental mutations:

• pax-3: linked to Waardenburg syndrome
• pax-6: linked to Aniridia
• pax-2: linked to Wilms tumor

During embryogenesis, Pax-2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Pax-2 mRNA and protein levels decrease as tubular epithelium matures. However, Pax-2 expression persists in the undifferentiated epithelium of Wilms’ tumors, correlating with proliferation of poorly differentiated epithelial cells. Expression of the Wilms’ tumor suppressor protein WT1 coincides with down-regulation of pax-2, suggesting WT1 may directly repress pax-2 transcription.
