Target Information
Oxygen radicals damage chromosomal DNA, causing cell death and mutations. While repair of 8-oxo-G has been a major focus, many other damaged DNA sites are generated by free-radical attacks. The human NTH1 repair protein acts on a wide range of these oxidatively damaged DNA sites. As a homologue of E. coli Endonuclease III, NTH is a DNA glycosylase with apurinic/apyrimidinic lyase activity. It exhibits broad substrate specificity, including numerous ring saturation and fragmentation products of pyrimidines. Research indicates that NTH plays a crucial role in removing oxidative base lesions in mitochondrial DNA.
