Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R), a type I glycoprotein (~75–80 kDa). It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding induces signaling through tyrosine kinase pathways, including Jak/STAT, essential for lymphocyte development and T-cell homeostasis. CD127 expression is notably down-regulated on CD4+CD25+ regulatory T cells (Tregs), serving as a marker for this population.

Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7. Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Tested: Pre-titrated; use 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Product # C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.

Target Information

CD127 (IL-7Rα) is a glycoprotein regulating lymphopoiesis. It forms an alpha/gamma chain heterodimer with IL7R, activating specific signaling pathways. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells. Dysfunction is associated with severe combined immunodeficiency (SCID) variants.
