Product Specific Information

The 17B5 monoclonal antibody reacts with mouse CD137 (4-1BB), a TNFR superfamily member expressed on activated T cells as monomers, dimers, and tetramers. CD137 interaction with its ligand 4-1BBL provides a costimulatory signal crucial for T cell and APC interaction. It may also be expressed on Tregs, influencing their proliferation. The 17B5 clone shows strong antagonistic effects in functional assays.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on stimulated mouse splenocytes at ≤ 0.5 µg/test (100 µL volume). Cell count range: 10⁵–10⁸ cells/test. Optimize antibody concentration empirically.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) with minimal impact on brightness and FRET efficiency.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD137 (TNFRSF9, 4-1BB) is an inducible costimulatory molecule expressed mainly on activated T cells. Its ligand, 4-1BBL, appears on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitors. CD137 signaling supports activated and memory CD8+ T cell survival, clonal expansion, and regulates cytokine release affecting monocyte survival. Soluble CD137 may act as a negative feedback regulator in immune responses.
