Description:
This 4B4 (4B4-1) monoclonal antibody reacts with human CD137 (also known as 4-1BB or TNFRSF9), an inducible member of the TNFR family expressed on T cells, NK cells, dendritic cells, granulocytes, and mast cells. CD137 signaling recruits TRAF1 and TRAF2, contributing to T cell activation, survival of activated and CD8+ memory T cells, and regulation of myelopoiesis and dendritic cell development. It also promotes expansion of CD4+CD25+ T regulatory cells ex vivo. The ligand 4-1BBL is found on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.

Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested on stimulated human peripheral blood cells.

• Recommended usage: 5 µL (0.125 µg) per test (in 100 µL final volume).
• Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the 488 nm blue laser.

• Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
• Ensure instrument compatibility before use.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:

Target Information

CD137 (4-1BB, TNFRSF9) is an inducible costimulatory molecule primarily expressed on activated T cells. Its ligand, 4-1BBL, is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells. CD137 signaling supports survival and expansion of activated and memory T cells, while regulating myelopoiesis and dendritic cell development. Soluble CD137 may act as a negative regulator in immune responses.
