Product Specific Information

The 13A4 monoclonal antibody recognizes mouse Prominin-1 (CD133), a 115–120 kDa pentaspan transmembrane glycoprotein. Prominin-1 is expressed on primitive cells such as hematopoietic, neural, endothelial stem cells, retina, and developing epithelium. The antibody does not cross-react with rat, human, chicken, or Drosophila antigens but has been reported to work in canine samples.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested with mouse bone marrow cells at ≤0.125 µg/test. One test = staining of a cell sample in 100 µL final volume (10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

PerCP-eFluor® 710 emits at 710 nm, excited by 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Fixation: Samples may be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

CD133 (AC133) is a 97 kDa glycoprotein with 5 transmembrane domains, appearing as a 120 kDa band due to glycosylation. It is a marker for immature hematopoietic stem cells and other progenitor cells including neural and embryonic stem cells. CD133 expression has been observed in various cancers (e.g., leukemias, brain tumors) and low levels in kidney, pancreas, placenta, and fetal liver. Mutations are linked to retinitis pigmentosa and Stargardt disease. Expression is regulated by multiple tissue-dependent promoters.
