Product Specific Information
The eBio500A2 monoclonal antibody reacts with the 25 kD epsilon subunit of the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells and NKT cells. Binding of eBio500A2 to CD3 initiates intracellular signaling leading to activation and proliferation. The 500A2 antibody partially cross-blocks the 17A2 and 145-2C11 antibodies, indicating recognition of distinct but overlapping epitopes.

Flow cytometry and immunohistochemical staining of frozen tissue sections.
Applications Tested:
Tested by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤ 0.25 µg per test (100 µL final volume). Optimal titration is advised for specific applications.

Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR complex expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related immunoglobulin superfamily members encoded on human chromosome 11. They associate with cytoplasmic signaling molecules through tyrosine-based motifs. CD3 participates in TCR-induced growth arrest, survival, and proliferation. CD3 antigen is found on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Reduced T lymphocyte levels may occur in autoimmune diseases. Defects in CD3 genes are linked to CD3 immunodeficiency.
