Product Specific Information
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. CD3e, along with CD3γ and CD3δ, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR triggers intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. It is commonly used as a phenotypic marker for mouse T cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse thymocytes and splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Cell numbers: 10⁵–10⁸ cells/test. Optimize titration for best performance.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded on human chromosome 11. Each subunit has cytoplasmic tails with tyrosine-based motifs for signal transduction. CD3 is present on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but absent on B and NK cells. CD3 gene defects are linked to CD3 immunodeficiency.
