Description:
The 1D11 monoclonal antibody reacts with human NKG2D, a 42 kDa lectin-like molecule expressed by NK cells, γδ T cells, CD8+ T cells, and some CD4+ T cells. Human NKG2D forms complexes with DAP10, a membrane adaptor protein, and can costimulate multiple NK activation receptors. The counter-receptor for NKG2D is MICA/MICB, expressed on epithelial tumors such as lung, breast, kidney, ovary, prostate, and colon carcinoma.
Both 5C6 and 1D11 block binding of soluble MICA to γδ TCR T cell clones and inhibit lysis by these cells. They also induce NKG2D-mediated redirected lysis of Fc receptor-bearing P815 cells.

For use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤ 0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Natural killer (NK) cells are lymphocytes capable of lysing tumor and virus-infected cells without prior activation. They regulate humoral and cell-mediated immunity and express several C-type lectins involved in NK cell function.
The NKG2 gene encodes a transmembrane protein with type II membrane orientation and a C-type lectin domain. The NKG2 gene family resides within the NK complex, which contains several lectin genes preferentially expressed in NK cells.
