Product Specific Information

Description:
The 28-14-8 monoclonal antibody reacts with mouse MHC class I, H-2D^b, and cross-reacts with H-2L^d, H-2D^q, and/or H-2L^q. Binding to H-2L^d alpha 3 domain is not dependent on beta2-microglobulin.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Validated by flow cytometric analysis of mouse splenocytes. Recommended ≤ 0.6 µg/test in 100 µL final volume. Cell number may vary (10^5–10^8 cells/test). Titrate for optimal performance.

Important Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL of CellBlox Plus per sample or per 100 µL of single-color control sample.

Technology:
NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures enabling engineered fluorescent signatures with minimized spillover and spread.

Emission: 756 nm
Laser: 633–640 nm (Red)

MHC class I alloantigen involved in antigen presentation to CD3/TCR and CD8-positive T cells.
HLA and MHC antibodies are key tools in immunopeptidomics, supporting neoantigen identification and characterization via LC-MS/MS.
