Product Specific Information

The 28-14-8 monoclonal antibody reacts with mouse MHC class I, H-2D^b, and cross-reacts with H-2L^d, H-2D^q, and/or H-2L^q. Binding to the alpha 3 domain of H-2L^d is independent of beta2-microglobulin.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes ≤ 1 µg/test (100 µL final volume, 10^5–10^8 cells/test). Optimize antibody titration for best performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer Usage:

  • Required for optimal staining with NovaFluor conjugates
  • Add 5 µL per sample before combining with cells
  • For single-color controls, use 5 µL per 100 µL of cell sample (10^3–10^8 cells)

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures, engineered for minimal spillover and spread.

Excitation: 552 nm
Emission: 663 nm
Laser: 561 nm (Yellow)

Internal testing shows that NovaFluor Yellow 660 non-specifically stains B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 mice. Other strains not tested.

Target Information

MHC class I alloantigen involved in antigen presentation to T cells expressing CD3/TCR and CD8.
HLA and MHC antibodies are useful in Immunopeptidomics for neoantigen identification via LC-MS/MS.
