Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in a wide variety of signal transduction pathways. Their levels are regulated by synthesis via nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs). PDEs form a superfamily of enzymes that convert 3′,5′-cyclic nucleotides to the corresponding nucleoside 5′-monophosphates.

PDE6 functions as the effector enzyme in the G protein-mediated visual signal transduction cascade. It consists of rod and cone-specific catalytic subunits (α′ for cone, α and β for rod), an inhibitory subunit γ, and a δ subunit of unknown function. The catalytic core comprises α′/α′ homodimers in cones and α/β heterodimers in rods. The C-termini of both catalytic and inhibitory subunits undergo methylation, myristoylation, and prenylation, which are essential for proper complex assembly and membrane association.
