Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in numerous signal transduction pathways. Their levels are controlled by synthesis via nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs). PDEs form a superfamily of enzymes that convert cyclic nucleotides (3′,5′-cAMP and cGMP) to their corresponding nucleoside monophosphates.

PDE6 serves as the effector enzyme in the G protein-mediated visual signal transduction cascade. It comprises five subunits: rod and cone-specific catalytic subunits (α′ for cone, α and β for rod), the inhibitory γ subunit, and δ subunit of unknown function. The catalytic core consists of α′/α′ homodimers in cones and α/β heterodimers in rods. The C-termini of both catalytic and inhibitory subunits undergo post-translational modifications (methylation, myristoylation, prenylation) essential for proper complex assembly and membrane association.
