Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in numerous signal transduction pathways. Their levels are controlled by synthesis through nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs). PDEs form a superfamily of enzymes that catalyze the conversion of cyclic nucleotides to their corresponding nucleoside monophosphates.

PDE6 functions as the effector enzyme in the G protein-mediated visual signal transduction cascade. It consists of five subunits: rod and cone-specific catalytic subunits (alpha', alpha, beta), an inhibitory subunit gamma, and a delta subunit of uncertain function. The catalytic core comprises alpha'/alpha' homodimers in cones and alpha/beta heterodimers in rods. Post-translational modifications such as methylation, myristoylation, and prenylation at the C-terminus are essential for proper complex assembly and membrane association.
