Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in diverse signal transduction pathways. Their levels are controlled by synthesis via nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs).
PDEs form a superfamily of enzymes that convert 3′,5′-cyclic nucleotides to nucleoside 5′-monophosphates.

PDE6 acts as the effector enzyme in the G protein-mediated visual signal transduction cascade. It comprises five subunits:

  • Catalytic subunits (α′ for cone, α and β for rod)
  • Inhibitory subunit (γ)
  • Delta subunit (δ) of unknown function

The catalytic core consists of α′/α′ homodimers (cone) and α/β heterodimers (rod). The C-terminus of both catalytic and inhibitory subunits undergoes methylation, myristoylation, and prenylation, essential for proper complex assembly and membrane association.
