Product Specific Information

This antibody detects endogenous levels of Caspase 9 only when phosphorylated at Threonine 125.

Target Information

Caspase 9 (also known as ICE-like apoptotic protease 6, ICE LAP6, Mch6, Apaf3) is a member of the peptidase family C14 containing a CARD domain. Caspases are cysteine proteases expressed as inactive precursors that mediate apoptosis via proteolysis of specific substrates.

Caspase 9 is active as a heterotetramer, localized in the cytosol, and translocates to mitochondria upon activation. It plays a central role in the caspase activation cascade responsible for apoptosis execution, cleaving and activating Caspase 3 and Caspase 6. Binding of Caspase 9 to Apaf-1 leads to activation of the protease, which then cleaves and activates Caspase 3.

Caspase 9 promotes DNA damage-induced apoptosis in an ABL1/c-Abl-dependent manner and cleaves poly(ADP-ribose) polymerase (PARP). It is expressed in various human tissues. In rats, an isoform with an alternative C-terminal sequence has been identified. Mutations in Caspase 9 affect brain and neuron development. In humans, tissue-specific expression variations are observed, and low Caspase 9 levels may be associated with cancer and neurodegenerative diseases such as Alzheimer’s disease.
