Product Specific Information
- Specificity: Recognizes the beta chain of the leukocyte integrin beta-2 family, including LFA-1, Mac-1, and p150.95.
- Epitope: Distinct from the C3bi binding site.
- Reactivity: Recognizes both human and Rhesus monkey antigens.
- Preparation: Blood from 4 healthy donors; mononuclear cells harvested and grown. BALB/c mouse immunized with monocytes, spleen cells fused to P3U1 myeloma cells to generate hybridomas. RIA used to identify lines of interest.
Target Information
CD18 (integrin beta-2 subunit) is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11a–d to constitute leukocyte (β2) integrins including:
- αLβ2 (CD11a/CD18, LFA-1)
- αMβ2 (CD11b/CD18, Mac-1, CR3)
- αXβ2 (CD11c/CD18)
- αDβ2 (CD11d/CD18)
These integrins mediate cell adhesion and signaling, essential for leukocyte migration and intercellular contact.
Deficiency or mutation in CD18 leads to leukocyte adhesion deficiency type I (LAD1) and related immune dysfunctions. CD18 also acts as a receptor for Mannheimia haemolytica leukotoxin, mediating cytolysis.
