Description:
The M17/4 monoclonal antibody reacts with mouse CD11a, a 180 kDa integrin alpha L, also known as lymphocyte function-associated antigen-1 (LFA-1) alpha chain. LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta 2), is an important adhesion molecule involved in lymphocyte and granulocyte function. CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3) are ligands for LFA-1. CD11a is expressed by all leukocytes.

Applications Reported:
This M17/4 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Can be used at ≤ 0.125 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimal titration is recommended for best performance.

eFluor® 450 is an alternative to Pacific Blue®, emits at 445 nm, and is excited with the Violet laser (405 nm). Ensure instrument compatibility.

Target Information

CD11a (LFA-1 alpha, ITGAL) with CD18 forms LFA-1 (αLβ2 integrin), crucial in leukocyte adhesion and activation. It binds ICAM-1, -2, and -3, mediating immune cell interactions and signaling. CD11a is expressed on leukocyte membranes, transitioning between low- and high-affinity states upon activation. It plays roles in immune cell trafficking, differentiation, proliferation, and T-cell effector functions. Blocking CD11a is a therapeutic approach for inflammatory diseases; for instance, Efalizumab (Raptiva) targets human CD11a. Simvastatin binding to CD11a also modulates immune activity. Two transcript variants encoding different isoforms are known. Disorders linked to CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
