Product Specific Information

The eBioKMC8 monoclonal antibody reacts with mouse CD9, a 24 kDa member of the transmembrane 4 superfamily. This family contains four hydrophobic domains spanning the cell membrane and short N- and C-terminal cytoplasmic domains.

CD9 is expressed by multiple cell types including:

  • Monocytes, macrophages, platelets
  • Early B cells, activated B and T cells
  • Dendritic cells, eosinophils, basophils
  • Endothelial cells, myoblasts, neuroblasts

On T cells, CD9 functions as a co-stimulatory molecule on naive T cells. CD9-deficiency can result in sterility due to defective gamete fusion. In macrophages, CD9 associates with FcgammaRs to modify phagocytosis and inflammatory signals. In B cells, CD9 is a marker for marginal zone B cells, B1 cells, and plasma cells. In dendritic cells, CD9 facilitates association of heterologous MHC II molecules. Expression levels vary by donor.

Applications Tested: Tested on mouse bone marrow cells; ≤1 µg/test recommended.
A test is defined as the amount of antibody staining a sample in 100 µL volume. Cell count range: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emits at 445 nm. Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.


Target Information

CD9 antigen is a glycoprotein expressed on developing B lymphocytes, platelets, monocytes, eosinophils, basophils, stimulated T lymphocytes, neurons, and glial cells. It belongs to the tetraspanin family, which spans the membrane four times.

In pre-B cells and platelets, CD9 regulates activation and aggregation via association with integrins CD41/CD61 (GPIIb/GPIIIa). CD9 is involved in:

  • Cell motility, osteoclastogenesis, neurite outgrowth, myotube formation, sperm-egg fusion
  • Cell attachment and proliferation
  • Association of MHC II molecules on dendritic cells for T cell stimulation

CD9 functions in differentiation, adhesion, and signal transduction, and plays a critical role in suppressing cancer cell motility and metastasis, acting as a metastasis suppressor in solid tumors.
