Product Specific Information
The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8α/β heterodimer or CD8α/α homodimer. CD8a is found on thymocytes, mature T cells, and NK cells. It binds to MHC class I and associates with p56lck, playing a key role in T-cell development and activation.
- Applications Reported: Flow cytometric analysis
- Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.25 µg) per test
- Fluorophore: eFluor® 450 (alternative to Pacific Blue®)
- Excitation: 405 nm
- Emission: 445 nm
- Laser: Violet
- Filtration: 0.2 µm post-manufacturing filtered
Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction and T-cell activation. CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer (CD8α/β) functioning as a co-receptor for MHC-I. It is also found as CD8α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide binding, CD8 recruits Lck kinase, triggering lymphokine production and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are crucial for pathogen and tumor clearance, with differentiation enhanced by IL-2, IL-12, and TGF-β1.
