Product Specific Information

The 10.1 monoclonal antibody reacts with human CD64 (FcRI), a 75 kDa type I transmembrane protein. CD64 is the high-affinity receptor for IgG and is involved in antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, and cytokine regulation. Monocytes and macrophages express CD64, while mature granulocytes and lymphocytes are negative.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells.
  • Recommended use: 5 µL (0.25 µg) per test
  • A test = antibody amount for 100 µL final volume
  • Cell count: 10⁵–10⁸ cells/test (empirically determined)

Super Bright 600 Characteristics:

• Tandem dye excited by violet laser (405 nm), emits at 600 nm
• Recommended filter: 610/20 bandpass
• Light sensitive — protect from light

Staining Panel Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation Compatibility:
Samples can be stored in:

• 1-step Fix/Lyse Solution (Product # 00-5333)
  for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

CD64 (FCGR1A) encodes a high-affinity Fc-gamma receptor involved in immune response regulation. It is one of three related genes on chromosome 1. CD64 dysfunction has been linked to cervical adenitis and tetrasomy 21.
