Heat-mediated antigen retrieval is recommended prior to staining using a 10 mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is lung tissue.

Target Information

iNOS (Inducible Nitric Oxide Synthase) produces nitric oxide (NO), a gaseous free radical involved in vasorelaxation, neurotransmission, and cytotoxicity.
NO production is mediated by NOS family enzymes: neuronal NOS (nNOS, type I), endothelial NOS (eNOS, type III), and inducible NOS (iNOS, type II).
iNOS is expressed in liver and inducible by lipopolysaccharide and cytokines such as IFN-γ, TNF, IL-1, and IL-2.
It is found in macrophages, hepatocytes, synoviocytes, and smooth muscle cells.
Cytokine induction increases iNOS mRNA, protein, and activity, resulting in sustained NO production.
iNOS dysfunction is associated with diseases such as achalasia and impotence.
