This ebic6 monoclonal antibody reacts with human Epstein-Barr Virus Induced Gene 3 (EBI3).
EBI3 may be secreted as monomers or homodimers, originally identified in virally infected B cells. It can also be secreted by trophoblast cells, potentially contributing to immune tolerance during pregnancy.

EBI3 is a component of two heterodimeric cytokines in the IL-12 family:

• IL-27 (EBI3 + p28): Secreted by antigen-presenting cells; exhibits both pro- and anti-inflammatory effects. It promotes Th1 differentiation and suppresses Th2/Th17 proliferation.
• IL-35 (EBI3 + p35): Produced by regulatory T cells; suppresses effector T-cell proliferation and induces regulatory phenotypes in CD4+ cells.

ELISA results from stimulated human dendritic cells suggest that some EBI3 detected by flow is dimerized with p28 to form IL-27. Testing in the context of IL-35 is ongoing.

Use at 5 µL (0.06 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test).

Target Information

EBI3 (Epstein-Barr virus Induced-3) is a secreted glycoprotein of the hematopoietin receptor family, regulating Th2-type immune responses and inflammation.
It dimerizes with p28 and p35 to form IL-27 and IL-35, respectively:

• IL-27: Produced upon TLR ligation by activated antigen-presenting cells; negatively regulates Th17 differentiation.
• IL-35: Involved in immunoregulation and suppression of effector T-cell responses.

EBI3 expression in dendritic cells is controlled by TLR signaling through MyD88 and NF-kappaB. It inhibits delayed-type hypersensitivity by suppressing IL-17 and inducing IL-10.
EBI3 may also regulate tumor metastasis via CD8+ T cells, with deficiency linked to reduced Th2 cytokine production and diminished allergic airway inflammation.
