Phosphoinositide-specific phospholipase C (PLC) plays a key role in receptor-mediated signal transduction by generating two second messengers, inositol 1,4,5-triphosphate and diacylglycerol, from phosphatidylinositol 4,5-bisphosphate.
There are multiple mammalian PLC isozymes including PLCβ1–4, PLCγ1–2, PLCδ1–2, and PLCε.
PLCδ isoforms, particularly PLCδ1, act as calcium signal amplifiers and are involved in oxytocin receptor and α1B-adrenoreceptor signaling. Abnormal accumulation of PLCδ has been observed in Alzheimer’s disease brains, suggesting a potential role in AD pathology.
PLCδ2 expression is associated with intestinal metaplasia and adenocarcinoma, serving as a potential marker for gastric malignant transformation.
