Target Information
TRAIL (TNFSF10) is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells, while sparing normal cells despite being expressed in most tissues. TRAIL binds to TNF receptor superfamily members such as TRAILR1 (TNFRSF10A), TRAILR2 (TNFRSF10B), TRAILR3 (TNFRSF10C), TRAILR4 (TNFRSF10D), and possibly OPG (TNFRSF11B).
Its activity is modulated by decoy receptors (TRAILR3, TRAILR4, OPG) that lack apoptosis-inducing capability. Binding of TRAIL to its receptors activates MAPK8/JNK, caspase 8, and caspase 3 pathways.
TRAIL is a type II membrane protein expressed in various human tissues and, similar to TNF and Fas ligand, induces apoptosis and NF-κB activation in many cells.
