Target Information

Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily and acts as a ligand for programmed death protein 1 (PD-1). It is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment.

PD-L2 expression is upregulated by Th2 cytokine IL-4 and to a lesser extent by Th1 cytokines TNF-α and IFN-γ. Although expressed at lower levels than PD-L1, PD-L2 has a 2–6 times higher affinity to PD-1. The PD-1/PD-L1/PD-L2 pathway serves as an inhibitory immune checkpoint, regulating T-cell activation and cytokine secretion (e.g., IFN-γ, IL-10).

Recent studies suggest PD-L2 also binds to a second receptor, RGMb (repulsive guidance molecule b), involved in respiratory tolerance via BMP and neogenin signaling pathways. The native human PD-L2 monomer comprises a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
