Target Information

Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for programmed death protein 1 (PD-1). PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment.

Expression is upregulated by cytokines such as IL-4, TNF-α, and IFN-γ. Although expressed at lower levels than PD-L1, PD-L2 has a 2–6× higher affinity for PD-1. The PD-1/PD-L1/PD-L2 pathway serves as an inhibitory immune checkpoint, regulating T-cell activation and cytokine secretion (e.g., IFN-γ, IL-10).

Recent studies also show PD-L2 interacts with RGMb (repulsive guidance molecule b), influencing respiratory tolerance through BMP and neogenin signaling. The native human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
