Product Specific Information
The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of CD22α and CD22β chains (130 and 140 kDa), produced by alternative splicing. It is highly expressed on mature B cells and B-cell lymphomas.
The extracellular region contains seven Ig-like domains capable of binding sialic acid moieties on epithelial, endothelial, B, and T cells. The intracellular region includes six tyrosine residues within ITIM and activation-like motifs, which are phosphorylated upon B-cell receptor engagement, enabling CD22 to regulate B-cell receptor signaling positively and negatively.
Applications Reported: Suitable for flow cytometric analysis and immunohistochemical staining.
Applications Tested: Verified by flow cytometric analysis of normal human peripheral blood cells at ≤0.5 µg/test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Titration recommended for optimal performance.
Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19, and is limited to late differentiation stages (mature B cells to plasma cells).
CD22 is useful in phenotyping mature leukemias and is strongly expressed in hairy cell leukemia. It binds preferentially to α2,6-linked sialic acid, with cis interactions potentially masking the binding site. Upon ligand-induced tyrosine phosphorylation, CD22 regulates B-cell receptor signaling through both activation (via Src kinases) and inhibition (via SH2 domain phosphatases).
CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
