Target Information
TLR5 is a Type I membrane protein belonging to the Toll-like receptor family and participates in the innate immune response to microbial agents. It mediates detection of bacterial flagellins and acts via MyD88 and TRAF6, leading to NF-κB activation, cytokine secretion, and inflammatory response. TLR5 binds to TIRAP and MyD88 via TIR domains and is highly expressed in liver, detected in lung, and at low levels in most other tissues. The TLR5 gene locus is associated with susceptibility to Salmonella infection. Mouse strains vary in resistance to S. typhimurium, and a common TLR5 stop codon polymorphism is linked to susceptibility to Legionnaires’ disease, highlighting the importance of TLR5 in innate immunity.
