Target Information

GAD-65 and GAD-67 belong to the group II decarboxylase family and catalyze the rate-limiting step in the production of GABA (gamma-aminobutyric acid) from L-glutamic acid.

• Both are found in the brain.
• GAD-65 localizes to synaptic vesicle membranes in nerve terminals.
• GAD-67 is distributed throughout the cell and responsible for basal GABA synthesis.
• During increased neurotransmission demand, GAD-65 transiently activates to assist in GABA production.
• Loss of GAD-65 impairs GABA neurotransmission; loss of GAD-67 is lethal.
• Two isoforms exist due to alternative splicing: GAD-67 (predominant) and GAD-25 (minor).
