Product Specific Information

The 10C4 monoclonal antibody reacts with mouse and rat Bcl-2, also known as B cell lymphoma 2. Bcl-2 is the founding member of the Bcl-2 family of proteins that regulate cell death and survival. Members include pro-apoptotic proteins (BAD, Bax, Bim) and anti-apoptotic proteins (Bcl-2, Bcl-xL, Mcl-1).
Bcl-2 inhibits apoptosis by preventing cytochrome c release from mitochondria. Mutation or overexpression of Bcl-2 due to chromosomal translocation contributes to various cancers.
In thymocytes, Bcl-2 is highly expressed in CD4–CD8– double negative and CD4+ or CD8+ single positive cells, but diminished in CD4+CD8+ double positive thymocytes.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis, immunoprecipitation, and immunoblotting (WB).
Fluorochrome-conjugated 10C4 is recommended for intracellular flow cytometry.

Applications Tested:
Validated for western blot at 1–10 µg/mL; titration recommended for optimal assay performance.

Target Information

BCL-2 regulates apoptosis, functioning to inhibit or promote cell death. Family members dimerize to modulate apoptosis.
Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak promote cytochrome c release, initiating apoptosis.
Constitutive expression of BCL-2 (e.g., via translocation to Ig heavy chain locus) is linked to follicular lymphoma.
In follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, while normal centers are negative.
Two transcript variants exist, differing at the C-terminal end. Overexpression of BCL-2 is associated with cancers such as B-cell lymphoma and prostate cancer.
